DHEA suppresses lipopolysaccharide (LPS)-induced TNF-alpha production in macrophages by blocking p38 MAPK phosphorylation and AP-1 activation in vitro. LPS-stimulated production of IL-1alpha in THP-1 cells, an IL-1alpha-dependent cell line, was also inhibited by DHEA. We also found that DHEA suppresses IL-2 production in the mixed lymphocyte reaction (MLR) between activated T cells. The inhibitory effect of DHEA on IL-2 production was also observed in the presence of DHEA-induced nuclear translocation of NF-kappaB and suppression of the transcriptional activity of the IL-2 promoter. These results suggest that DHEA inhibits the cytokine production in human T cells and that the suppressive effect of DHEA on the MLR is partially mediated by suppression of IL-2 production by T cells. In addition, we found that DHEA suppresses the LPS-induced production of TNF-alpha in THP-1 cells and that the inhibitory effect of DHEA on the TNF-alpha production in THP-1 cells is related to the inhibition of IL-1alpha production in THP-1 cells.
